一种基于蛋白基水凝胶体外构建神经母细胞瘤微组织的模型的构建方法及其药物筛选方法,为解决NB异质性高、个体化差异大而带来的难治性问题提供了技术支持,为NB的精准化和个性化治疗提供技术解决方案,本发明以明胶衍生物—甲基丙烯酸化明胶(GelMA)作为三维细胞培养的支架,通过固含量的改变调控支架的机械性能,将其作为恶性分化程度较高的神经母细胞瘤细胞 SH-SY5Y 的三维培养载体,观察肿瘤细胞在不同于体内的存活、增殖以及肿瘤球状体形成的规律;将形成的三维细胞肿瘤模型进行体外肿瘤化疗药物共培养,观察其对化疗药物的敏感性,为神经母细胞瘤的药物筛选和精准治疗提供一种新的技术平台。

GelMA hydrogel with different concentrations were prepared as 3D scaffolds for NB cell line SH-SY5Y. Cell proliferation, migration in the hydrogel scaffold, and the process of tumor organoids formation were observed. Chemotherapeutic sensitivity of tumor suppressor was tested by co-culture with tumor organoids. In summary, the tumor organoids constructed in this study provides a promising option for establishing an effective anti-tumor drug screening platform.
